My brother is separated from his wife and he has a family vehicle that is in his name and he made all the payments (she doesn't work). She calls him and will make statements like "give me MY car back" trying to trick him into saying it is hers, so she can use it in court. What holds up in court?

The car is his until a judge says otherwise.

However... if she has her own keys and is able to drive it away, it becomes hers until a judge says otherwise
If they are married it belongs to both of them... doesn't matter who is working or who paid for it.
